abstract | The invention discloses a preparation method of a nucleus pulposus cell modified porcine small intestine submucosa multiple acellular material, which comprises the steps of stripping porcine SIS tissues, treating the stripped porcine SIS tissues by a physiological saline buffer solution containing a protease inhibitor, an organic solvent solution, a PBS buffer solution containing Triton X, a PBS buffer solution containing SDS and a PBS buffer solution containing DNase to obtain the acellular SIS materials, carrying out ball milling on the acellular SIS materials to obtain particles with the diameter of 200 mu m, co-culturing the particles with separated nucleus pulposus cells and carrying out acellular treatment again to obtain the multiple acellular injectable nucleus pulposus repairing material. The invention can keep the integrity of the original ECM while removing foreign body or foreign cell with immunogenicity, has good extracellular microenvironment, biochemical factor, biomechanical property and the like, and can simulate the growth environment of nucleus pulposus cells in normal physiological condition to the maximum extent. |
