abstract | The invention relates to the field of biotechnology, and particularly discloses a primer, a probe combination and a detection method for rapidly detecting M gene in Porcine Epidemic Diarrhea Virus (PEDV) by using RPA technology. The sequence of the forward primer is shown as SEQ ID No.1, the sequence of the reverse primer is shown as SEQ ID No.2, and the sequence of the probe is shown as SEQ ID No. 3. The RPA method established by the invention can react for 20min at the constant temperature of 39 ℃, can realize effective detection of PEDV, takes puc57 plasmid containing PEDV virus M gene fragment as a template, and has the RPA detection limit of 10 1 Copying; RPA specifically amplifies only the gene of PEDV virus M, but not the gene sequences of the membrane protein or nucleocapsid protein of TGEV, PRRSV, PCV-2 and CSF viruses. Furthermore, the RPA method can be used for effectively detecting the PEDV nucleic acid in the plasma and the plasma protein powder. The RPA method established by the invention has the advantages of simple operation, quick response, accurate detection and high sensitivity, and provides a new and reliable technical support for the detection, prevention and control of PEDV. |
