abstract | The invention discloses a 3, 7-dihydroxy tropolone biosynthesis gene cluster and application thereof. The biosynthetic gene cluster comprises 8 structural genes: trlA, trlB, trlC, trlD, trlE, trlF, trlH, and trlI; 1 regulatory-related gene: trlJ; and 1 other functional gene: trlG. Wherein trlB and trlJ are key regulatory proteins participating in strengthening the metabolic pathway of the chorismate, and trlC, trlD and trlE provide tool enzymes capable of performing sequential oxidation on the tropolone seven-membered aromatic ring. The gene and the protein thereof provided by the invention can be used for discovering or researching new compounds, genes or proteins for medicine and industry and agriculture. The invention also provides a series of engineered strains capable of producing different tropolone compounds such as tropolone, 7-hydroxytripolone, tropone and 1,4, 6-tropone-1-carboxylic acid in high yield. |
