abstract | The invention discloses an estrol hapten, a complete antigen and a preparation method thereof. The method comprises the following steps: reacting the estrol and monochloroacetate in an organic solvent at normal temperature in the presence of a catalyst, stopping the reaction by ice water, acidifying the water phase of the unreacted estrol in an organic extractant by acid to generate white precipitate, and thus obtaining the estrol hapten. The method provides a simple method without heating and controlling pH through optimizing a catalyst, reaction time and the like in synthesis, and the hapten can be obtained in a short time; the method is characterized in that the product and the substrate have different dissolution characteristics, unreacted estrol substances are removed by an extraction method, and meanwhile, the obtained estrol substances can be continuously used by recovery, so that the reaction substrate is saved; finally, the high-purity estrol hapten can be obtained by a crystallization method through a hydrochloric acid acidification method; the complete antigen is obtained by connecting the estrol hapten and carrier protein. |
