abstract | The patent application discloses an acegastrodine derivative and a preparation method thereof, wherein the acegastrodine derivative is a derivative with a general formula I:n wherein R represents alkyl or alcohol compounds, and the preparation method comprises the following steps: firstly, maleic anhydride and furan are subjected to Diels-Alder reaction at 38 ℃ by taking ether as a solvent to prepare 5-alkene norcantharidin, then substitution reaction of various alcohol reagents on anhydride is carried out, finally 4- (dimethylamino) pyridine (DMAP) is used as a catalyst, 1-ethyl- (3-dimethylaminopropyl) carbonyldiimine hydrochloride (EDCi) is used as a condensing agent, and the temperature is controlled to be 0-55 ℃ to carry out condensation reaction with acetyl gastrodin; the method has the advantages of simple process, low requirements on equipment and conditions, easily obtained raw materials, high yield and purity, less discharge of three wastes and suitability for industrial production. The obtained acetyl gastrodine derivative has anti-liver cancer and anti-tumor activity. |
