abstract |
The present invention provides a cell line that reduces the production of replicable adenovirus and its construction method and application. The cell line HEK293.CS that reduces the production of replicable adenovirus is obtained by knocking out the gene fragment homologous to Ad5 adenovirus E1 gene in HEK293 , while providing a template plasmid to replace it with a non-homologous sequence that can stabilize the expression of the E1 gene, a safe adenovirus production cell line was constructed. Compared with the unmodified HEK293 cell line, the growth ability and production capacity of HEK293.CS The virus ability is not reduced, but no detectable RCA is produced; the cell line can be used for large-scale culture of recombinant human adenovirus type 5, and reduces the probability of RCA production in the process of manufacturing vaccines, antibodies and other drugs. |
