The present invention provides lipid A mimetics in which one or two sugar residues in the native lipid A disaccharide backbone have been replaced by aryl groups. These lipid A mimetics may further differ from native lipid A molecules in other structural features, such as the presence of different numbers of phosphate groups, changes in the number, structure and position of lipid chains and/or sugar residues (or their Aromatic substitutions) steric and linkage changes. A lipid A mimetic may be a lipid A agonist and thus useful as an immunostimulatory agent to induce or permit antibody and/or cell-mediated immune responses, or may be a lipid A antagonist and thus useful in therapy or prophylaxis Lipopolysaccharide (LPS)/Lipid A mediated disease or disorder. Also provided are methods of making the lipid A mimetics. |
