abstract | The invention discloses a culture method of Schwann cells, which comprises the following steps: s1, culturing the cord blood mononuclear cells by using a MSCs culture medium to obtain P1 generation cord blood MSCs; s2, culturing the P1 generation cord blood MSCs in a neurosphere culture medium to obtain P3 generation neurospheres; and S3. culturing the P3 generation neurosphere in a Schwann cell culture medium to obtain Schwann cells. The invention has the beneficial effects that: the culture method of the Schwann cells is a method for culturing the Schwann cells derived from cord blood MSCs through a nestin + neurosphere stage. The P1 generation cord blood MSCs are adopted to induce the formation of neurospheres, and the neurosphere formation rate is as high as 41%. The invention also provides a specific neurosphere culture medium which is enough to induce the MSCs of the umbilical cord blood to form nestin + neurospheres; provides a specific culture scheme for differentiation from nestin + neural precursor cells to schwann cells, can remarkably promote the attachment and migration of nestin + neural precursor cells, and prevent agglomeration and multilayer growth in the cell proliferation process. |
