abstract | The invention discloses a method for preparing and cryopreserving fat SVF cells clinically and efficiently and an application thereof, wherein the preparation method comprises the following steps: step S1: collecting a fat sample under an aseptic condition, storing the fat sample in a preservation solution, and adding a washing solution to pretreat the fat sample; step S2: centrifuging the pretreated fat sample to obtain yellow fat tissue, taking out the fat tissue, adding equal volume of preheated digestive juice, sealing, uniformly mixing the fat tissue and the digestive juice at constant temperature, digesting until no large tissue mass exists or a large amount of lipid is accumulated although the large tissue mass exists, and centrifuging; step S3: and (3) after the centrifugation in the step 2 is finished, removing the upper-layer lipid, adding a washing solution to resuspend and uniformly mix the precipitate, filtering, adding the washing solution again, centrifuging, and removing the supernatant to obtain the fat SVF cells. The washing liquid and the digestive liquid adopted by the preparation method greatly increase the cell number, the cell viability and the purity of the prepared fat SVF cells, and ensure the effect of the extracted SVF cells in clinical treatment. |
