abstract | The invention discloses a method for amplifying NK cells without trophoblasts, which comprises the following steps: by blendingCulturing NK cells in a culture solution for 7 days; then, culturing with autologous plasma culture medium, supplementing liquid every 2-3 days during the culture process, and maintaining the cell density at 2.0 × 10 6 mL, culturing for more than 20 days; the composition of the blended culture solution is as follows: adding 1-10% of autologous plasma, 200-1500 IU/mL of IL-2, 10-50 ng/mL of IL-15 and 10-50 ng/mL of IL-21 on the basis of a lymphocyte culture medium; the composition of the autologous plasma culture medium is as follows: on the basis of a serum-free culture medium, 1-10% of autologous plasma and 200IU/mL of IL-2 are added. According to the method for amplifying the NK cells without the feeder cells, the cord blood or the autologous peripheral blood is used as a cell source, the feeder cells are not needed in the culture process, the amplification efficiency is high, and the purity of the obtained NK cells is high. The NK cells obtained by the method can be used as a pharmaceutical composition for cellular immunotherapy, and are mainly used for treating and/or preventing infectious diseases and/or cancers. |
