abstract The invention relates to a chimeric antigen receptor, an expression gene thereof, a T cell modified by the chimeric antigen receptor and light-operated regulation and application thereof. The chimeric antigen receptor expression gene includes a first fusion protein expression gene and a second fusion protein expression gene. The first fusion protein expression gene comprises an antigen binding region expression gene, a hinge protein expression gene, a first transmembrane protein expression gene and an arabidopsis thaliana cryptochrome protein-2 expression gene which are sequentially connected. The second fusion protein expression gene comprises a second transmembrane protein expression gene, an alkaline helical loop helical protein-1 expression gene and an intracellular signal transduction protein expression gene which are sequentially connected. The chimeric antigen receptor expression gene with the innovative design can be successfully introduced into T cells to form chimeric antigen receptor modified T cells, and immunoreaction can be generated under the specific blue light condition and the specific antigen condition, so that the chimeric antigen receptor immunoreaction is controllable, the treatment side effect is less, and the specificity is high.
