The present disclosure provides a method for preparing a sequencing library comprising the steps of providing a template nucleic acid sequence, dNTP, dUTP, primers, polymerase, dUTP excision enzyme, and a plurality of oligonucleotide adapter sequence segments comprising beads; utilizing the polymerase, dNTP, dUTP and random hexamers to amplify the template nucleic acid to provide complementary nucleic acid sequences comprising occasional dUTPs; and utilizing the dUTP excision enzyme to excise incorporated dUTP to nicks are provided in said complementary nucleic acid sequence, thereby providing a sequencing library. |
