abstract | The invention relates to a method for chiral preparation of (S) -tetrahydro-1-naphthoic acid shown in formula (V) and derivatives thereof, which comprises the following steps: (a) naphthoic acid and its derivative shown in formula (I) and (II) R ) The-2-hydroxy carboxylic ester is subjected to esterification reaction to generate naphthoate shown in the formula (III) and derivatives thereof. (b) The naphthoic acid ester shown in the formula (III) and the derivative thereof are subjected to hydrogenation reduction reaction in a proper solvent under the action of a metal catalyst to obtain (IV) S ) -1,2,3, 4-tetrahydronaphthalene-1-carboxylic acid esters and derivatives thereof. (c) Formula (IV) shown in (A) S ) Hydrolysis of (E) -1,2,3, 4-tetrahydronaphthalene-1-carboxylic ester and derivatives thereof under basic conditions to give (A) S ) -1,2,3, 4-tetrahydro-1-naphthoic acid and derivatives thereof. The invention provides a chiral preparation (different from the prior art), which has the advantages of simple operation, environmental protection and high atom economy and is suitable for industrial production S ) -1,2,3, 4-tetrahydro-1-naphthoic acid and derivatives thereof. |
