abstract | The invention discloses a method for separating human placental chorionic mesenchymal stem cells, which comprises the following steps: 1) shearing healthy human placenta chorion tissue into strips and fully washing with normal saline; 2) adding appropriate amount of normal saline, cutting chorion into fine particles with homogenizer, and washing with normal saline to clarify; 3) processing the mixture to be micro-particles by using a homogenizer again, and centrifuging to remove upper layer blood cells; 4) adding enzyme into the precipitate for digestion, and centrifugally separating the precipitate; 5) after centrifugal washing twice by normal saline, adding a culture medium into the sediment for inoculation and culture; 6) and on the 7 th day, removing tissue blocks and culture solution, flushing the bottom of the culture flask with normal saline, adding a fresh culture medium, replacing the culture medium every 3 days until the cell fusion degree reaches 80-90%, and digesting and passaging with pancreatin to obtain the human placental chorionic mesenchymal stem cells. The method is simple, convenient and quick to operate, and the high-quality human placental chorionic mesenchymal stem cells can be obtained. |
