abstract | The invention discloses a method for preparing the protoplast of Scenedesmus obliquus. The algal liquid of Scenedesmus obliquus cultivated to the logarithmic growth phase is collected, oscillated with an ultrasonic oscillator, centrifuged to collect the Scenedesmus obliquus cells, and regenerated with BG11 medium. Suspended to obtain the cell suspension of Scenedesmus obliquus; using the mixed digestive enzyme solution extracted from the large flea, adding a small amount of cellulase, pectinase and hypertonic solvent mannitol and membrane stabilizer CaCl 2 , and suspended with Scenedesmus obliquus cells. The solution is mixed, and the cell wall of Scenedesmus obliquus is digested by enzymolysis to obtain protoplasts. The mixed Digestive Enzyme activity of Flea daphnia applied in the method of the invention is high, and the cell wall of Scenedesmus obliquus can be enzymatically hydrolyzed under mild conditions in a short time. The efficiency of preparing protoplasts by this method can reach 70%, and the protoplasts have strong activity and regeneration ability. The invention provides technical support for cell fusion and gene manipulation targeting Scenedesmus obliquus, and also provides reliable raw materials for studying the endocytosis of microalgal cells. |
