abstract | The invention discloses a method for fusing potato tetraploid cultivars and diploid wild species protoplasts, which is characterized in that a set of optimal conditions for separating and purifying test-involved potato protoplasm, a protoplast culture medium, a callus regeneration culture medium and a tetraploid and diploid protoplast fusion method are obtained by searching and optimizing a set of protoplasts obtained by separating and purifying potato protoplasm by adopting an enzymolysis method and an interface method and fusing the tetraploid and diploid protoplasts by adopting a polyethylene glycol (PEG) induction fusion method. In a fusion experiment of Fiurodebuit and wild diploid materials (E12 and E25) carried out by using the method, 2 fusion strains and 4 fusion calluses which are detected by SRAP molecular markers through DNA are obtained; these materials can be used as subsequent experimental materials to determine the variation of the agronomic traits and the possible applications in production of the obtained fusion lines. |
