abstract | The invention discloses a purification method of nostoc sphaeroides hemoglobin, which comprises the following steps: (1) selecting an anion exchange column to pass through the column; (2) preparing a PB Buffer solution Buffer A and a PB-NaCl Buffer solution Buffer B; (3) dissolving the crude powder of the nostoc sphaeroides phycobiliprotein with PB Buffer solution Buffer A, centrifuging and filtering; (4) washing a pump, and balancing an ion exchange column by using a PB Buffer solution Buffer A; (5) directly sampling nostoc sphaeroides biliprotein dissolved in PB Buffer solution Buffer A prepared in the step (3), carrying out gradient elution by using PB-NaCl Buffer solution Buffer B, and collecting eluent; (6) washing the ion exchange column with a saline solution, and then back-flushing with an alkaline solution; (7) ultrafiltering, concentrating, and dialyzing to obtain purified water solution of nostoc sphaeroides hemoglobin. The method of the invention has simple operation, and the concentration and purity of the obtained nostoc sphaeroides hemoglobin are obviously improved, thus being suitable for industrialized large-scale production. |
