abstract | The invention discloses a construction method of a reporter gene cell strain based on endoplasmic reticulum stress, which comprises the steps of selecting CHOP genes related to endoplasmic reticulum stress as specific induction genes to construct CHOP promoters, and connecting the CHOP promoters with SEAP genes convenient to detect to construct a lentiviral CHOP-SEAP plasmid vector; finally, the CHOP-SEAP plasmid was transfected into HeLa cells. The invention also discloses application of the reporter gene cell strain constructed by the construction method based on endoplasmic reticulum stress. The reporter gene cell strain constructed by the method can indicate early cytotoxic damage, greatly improve the detection sensitivity of the cell strain and reduce the detection limit value, thereby representing the early toxicity of toxic pollutants and being suitable for most toxic pollutants; meanwhile, the comprehensive toxicity of the actual water body can be represented. The reporter gene SEAP gene selected by the cell strain has the advantages of convenient detection and dynamic monitoring. |
