abstract | The invention relates to a method for preserving donkey semen at low temperature, which comprises the following steps: (1) preparing base liquid by 3.04 parts of Tris, 1.70 parts of citric acid, 1.50 parts of glucose, 1.50 parts of fructose, 0.06 part of penicillin, 0.10 part of streptomycin and 100 parts of double distilled water, and preparing semen diluent by using 85 parts of the base liquid and 15 parts of yolk liquid; (2) taking an estrus female donkey as a donkey, and collecting semen of a male donkey by a pseudo-vaginal method, wherein the survival rate is more than 0.70; (3) taking semen diluent and semen with the temperature balanced to 28 ℃, and diluting according to the proportion of 2: 1; (4) placing the diluted semen into a centrifuge tube, placing the centrifuge tube in a water bath at 28 ℃, and transferring the centrifuge tube into a refrigerator at 4 ℃; (5) the mixture is uniformly mixed every 24 hours during low-temperature storage, and the sperm motility rate is examined under a microscope. The semen preservation time is obviously prolonged after the method is adopted, the difficulty of mass artificial insemination in a large-scale donkey breeding field when the female donkey is in concentrated estrus is solved, and the utilization efficiency of male and male donkeys is effectively improved. |
