abstract The invention relates to a method for prolonging truncated protein of nonsense mutant of pathogenic gene by constructing suppressive tRNA, so that full-length functional protein is produced in mammalian cells, thereby restoring normal structure and function of the mutant. The system related to nonsense mutation in the invention comprises a pathogenic gene of monogenic genetic diseases and a tumor suppressor gene in tumor cells. The invention mainly relates to the construction of 19 suppressor tRNA corresponding to three stop codons, which can efficiently read dystrophin protein in mammalian cells and read nonsense mutant protein in tumor cells, and has obvious effect.
