abstract | The invention relates to a gene detection method, in particular to a kit for detecting single nucleotide polymorphism and a detection method; determining a detection site of a gene to be detected; respectively designing a forward primer and a reverse primer aiming at each detection site; in the primers corresponding to the two adjacent sites, complementary base sequences are arranged at the 5 end of the reverse primer of the upstream site and the 5 end of the forward primer of the downstream site; performing first PCR amplification by using DNA of a gene to be detected as a template and using a primer to obtain a first PCR product; taking the mixture of the first PCR products as a template, taking a forward primer corresponding to the most upstream site and a reverse primer corresponding to the most downstream site as a primer pair, and carrying out second PCR amplification to obtain a second PCR product; sequencing the second PCR product to complete the detection of single nucleotide polymorphism; through the first-generation sequencing detection, the mutation information of a plurality of gene loci can be obtained simultaneously, and the method is a technology for rapidly detecting the single nucleotide polymorphism. |
