abstract The invention provides a method for improving induction efficiency and genetic transformation efficiency of hairy roots of bupleurum chinense and application thereof. The optimal combination condition for improving the infection efficiency of the agrobacterium rhizogenes is confirmed based on the exploration of the agrobacterium rhizogenes strain, the OD value of the infection bacterial liquid, the co-culture time and other conditions, and compared with the traditional hairy root induction rate of about 10%, the hairy root induction rate obtained by the technical scheme provided by the invention reaches 70% -85%. The method for improving the genetic transformation efficiency of the hairy roots of bupleurum chinense can obtain the transformation rate of more than 80 percent of exogenous genes, and provides better, more efficient and simpler selection for researching the functional genes of bupleurum chinense and/or industrialization of metabolites of the hairy roots of bupleurum chinense.
