The present invention relates to novel methods of characterizing a target RNA polynucleotide by taking one or more measurements as the target RNA polynucleotide moves relative to a transmembrane pore. This movement is controlled by DNA helicase. The invention also relates to modified RNA constructs, wherein the RNA polynucleotide has been modified to enhance binding thereto by a DNA helicase. |
