abstract | The present invention provides methods for deriving adenohypophysis or precursor tissue thereof from human pluripotent stem cells in vitro. In this method: a hypothalamic neuroepithelial tissue and epidermal ectoderm are obtained by culturing human pluripotent stem cell aggregates in suspension in a medium containing bone morphogenetic factor signaling pathway activators and Shh signaling pathway active substances of human cell aggregates; and by subjecting the resulting human cell aggregates comprising hypothalamic neuroepithelial tissue and epidermal ectoderm in a medium containing bone morphogenetic factor signaling pathway activators and Shh signaling pathway active substances Further suspension culture induces the formation of pituitary placodes and/or latke sacs in the epidermal ectoderm, resulting in human cell aggregates comprising 1) hypothalamic neuroepithelial tissue and 2) pituitary placodes and/or latke sacs. |
