abstract | The invention discloses a biosensing detection method for identifying salmon and trout fries, which is characterized in that 6 specific primers using IHNV virus matrix protein (M) coding genes as amplification targets are designed based on the conservative analysis results of IHNV gene sequences from different strains in a public database, and the total RNA template of a fish egg or fry sample to be detected is subjected to constant-temperature reverse transcription amplification, wherein the amplification time is only 15-30 min. All or part of primers in the amplification reaction carry electrochemical active markers, and part of dNTPs carry anchoring markers, so that the successfully amplified virus M gene sequence simultaneously carries the electrochemical active markers and the anchoring markers, an amplification product can be captured on the surface of an electrode by using the anchoring markers, and whether a sample carries IHNV is accurately judged by detecting electrochemical signals on the surface of the electrode. The method is simple to operate, quick in response, low in requirement on equipment precision, and suitable for field quick detection of production and management places such as fingerling farms and breeding farms. |
