abstract | The invention relates to a targeted fusion protein for resisting HBV replication and a construction method thereof. The peptide fragment is used as a guide molecule of targeting cccDNA and is marked as HBc144 by utilizing the characteristic that the C-terminal region (CTD) of the hepatitis B virus core protein HBc can be specifically combined with HBV cccDNA; synthesizing an A3A-HBc144 coding gene sequence and an HBc144-UNG2 coding gene sequence, cloning the two sequences to multi-cloning sites MCS1 and MCS2 of a eukaryotic expression vector pVITRO2-neo-MCS to construct a recombinant dual expression vector Pvitro2-A3A-HBc/HBc-UNG2, wherein the recombinant vector can simultaneously express two fusion proteins, namely A3A-HBc144 and HBc144-UNG2, in liver cells, the invention realizes the targeting of A3A and UNG2 to HBV cccDNA, and has stronger effect of inhibiting virus replication compared with a vector which independently expresses A3A or A3A-HBc. |
