abstract A new technology for direct cloning of large genomic fragments and DNA multi-molecule assembly. The present invention provides a method for homologous recombination, the method comprising treating two or more target nucleic acid molecules with a first exonuclease, and then allowing the treated target nucleic acid molecules to bind to a second exonuclease and an annealing protein. Homologous recombination is carried out in the presence of the target nucleic acid molecules in which homologous recombination occurs, sharing at least a sequence homology region. The invention also provides kits for use in the methods of the invention.
