abstract | The sialic acid-carboxylic acid compound conjugate is obtained by direct glycosidation reaction of a sialic acid thioglycoside donor and a carboxylic acid compound, and is a compound with sialic acid and carboxylic acid ester at the anomeric position, the preparation method comprises ⑴ steps of sequentially adding the sialic acid thioglycoside donor, a newly activated molecular sieve and the carboxylic acid compound into a reaction vessel, adding redistilled dichloromethane for dissolving, argon protecting, stirring for 15 minutes at 25 ℃, ⑵ steps of rapidly adding boron trifluoride diethyl etherate and iodosuccinimide into the activation system, stirring and reacting for 5-10 minutes at 25 ℃, ⑶ steps of filtering the reaction system, diluting the reaction system with an organic solvent, washing the organic phase once with a saturated sodium thiosulfate solution, extracting the aqueous phase with the organic solvent for three times, combining the organic phase, drying with anhydrous sodium sulfate, filtering, spin-drying, and eluting by silica gel column chromatography to obtain the sialic acid-carboxylic acid compound conjugate. |
