abstract | The invention aims to provide a method for detecting protein by using amino acid analysis. The method comprises the following specific steps: (1) weighing a sample in a hydrolysis tube, adding a p-phenolsulfonic acid hydrolysis reagent of an ethanol solution, ultrasonically mixing the sample uniformly, introducing argon or nitrogen to remove dissolved oxygen, sealing, and putting the sample in a vacuum drying oven for hydrolysis; (2) taking out the test tube hydrolysis tube, cooling to room temperature, opening the tube, filtering to a 50ml volumetric flask, washing and collecting washing liquid, and fixing the volume to the scale mark; (3) sucking 0.5-2ml of the sample with constant volume into a test tube, and completely drying the sample in a vacuum drying oven; (4) adding a sample buffer solution into a sample hydrolysate test tube after evaporation, placing the sample buffer solution on a vibration mixer for uniform mixing, sucking a small amount of the sample buffer solution by using a needle tube, filtering the sample by using a filter, performing amino acid analysis on the hydrolysate by using an amino acid analyzer, comparing an amino acid map of the obtained sample with a spectrogram of a standard sample, and calculating the type and the content of the amino acid in the sample according to the retention time and the peak area of each peak on the map.
