abstract | The invention discloses bifunctional glucanase with chitosan hydrolytic activity, a gene, a carrier, engineering bacteria and application thereof in chitosan degradation and chitosan oligosaccharide production, wherein the amino acid sequence of the bifunctional glucanase is shown in SEQ ID NO. 1. The novel high-temperature resistant bifunctional glucanase has good heat resistance, the optimal action temperature is 55 ℃, the enzyme activity is stable when the chitosan substrate is hydrolyzed under the condition of pH 2.0-6.5, 100g/L of high-concentration chitosan substrate can be efficiently hydrolyzed, the substrate is completely hydrolyzed within 8 hours, the product is low-molecular-weight chitosan oligosaccharide with the molecular weight below 1500Da, and no monosaccharide byproduct is generated. The enzyme activity can reach 1000U/ml, which is much higher than the general production level (10-100U/ml) of the prior chitosan hydrolase. |
