abstract The invention provides a specific sequence and a method for rapidly and accurately detecting the purity of melon seeds. The specific sequence is selected from one or more than two of the following sequence groups: SEQ ID NO.01 and SEQ ID NO. 02; SEQ ID NO.03 and SEQ ID NO. 04; up to SEQ ID NO.79 and SEQ ID NO. 80. The invention also provides a method for detecting the purity of the melon seeds, which comprises the following steps: (1) extracting sample genome DNA; (2) carrying out PCR amplification and detection on the sample mitochondrial genome DNA by using a primer sequence; (3) carrying out SSR polymorphism marking and characteristic strip statistics; (4) and calculating the purity of the melon seeds. The invention develops a set of melon mitochondrial genome SSR markers with high polymorphism for the rapid detection and identification of melon seed purity based on the parental genetic characteristic of melon mitochondrial genome for the first time.
