abstract The invention discloses an NK cell in-vitro amplification method which comprises the steps of (1) preparing a reagent A with a main component of 50ng/ml anti-CD 3 monoclonal antibody +50ng/ml anti-CD 16 monoclonal antibody, a reagent B with a main component of 300IU/ml I L-2 and a reagent C with a main component of 20ng/ml I L-12 +20ng/ml I L-15, preparing 40ml of lymphocyte separation liquid as a reagent D, (2) coating the reagent A in a T-175 culture flask, standing overnight at 4 ℃), treating peripheral blood to obtain cells and serum, (4) resuspending the cells, adding the reagent D and then centrifugally collecting the PBMC, (5) resuspending the PBMC in an X-VO VI 15 culture medium, adding the reagent B, the reagent C and autologous serum, incubating, and (6) supplementing the culture medium, the reagent B, the reagent C and the autologous serum on days 3, 6, 9, 12 respectively, collecting the cells on day 14, the benefit lies in that the anti-CD 3 and anti-CD 16 monoclonal antibody are inoculated in advance, and the purity of the cells is improved when the in-vitro culture is firstly.
