abstract | The invention discloses a multi-chamber microfluidic PCR chip for screening an adapter, which comprises a microfluidic PCR reaction plate, a temperature circulating heating plate, a glass substrate and a chip clamp, wherein a reaction liquid injection hole, a Z-shaped mixing unit, an inflow microfluidic channel, a PCR amplification chamber, an outflow microfluidic channel and a reaction liquid outlet hole are sequentially communicated on the PDMS reaction plate. The microfluidic PCR chip adopts a plurality of reaction chambers, so that the high quality, sufficient acquisition and nucleic acid variety diversity of a secondary library can be ensured; the microfluidic PCR chip can rapidly obtain the optimal amplification condition through a fluorescent real-time amplification curve, and amplify the nucleic acid sequence screened by the aptamer under the optimal condition. The invention also discloses a method for screening the aptamers by using the multi-chamber microfluidic PCR chip, which can be used for PCR amplification of screening of various target aptamers and provides good idea design and reference for other aptamer screening workers. |
