abstract The invention discloses a primer and a method for amplifying a satellite-head woodpecker magnetic induction receptor protein gene ISCA1 fragment, wherein the primer comprises a first primer pair and a nested PCR primer, and the first primer pair comprises PIC-2L-F with a sequence number of SEQ ID No.1 and PIC-2L-R with a sequence number of SEQ ID No. 7. The method comprises the following steps: (1) extracting total genome DNA of a sample to be detected; (2) carrying out PCR amplification on the total genomic DNA by adopting a first primer pair; (3) performing secondary amplification on the product amplified in the step (2) by using a nested PCR primer; (4) and (4) detecting the product obtained after the secondary amplification in the step (3). The invention can accurately, simply and rapidly obtain the long fragment sequence of the star-head woodpecker ISCA1 gene.
