abstract The invention discloses an ultralow temperature cryopreservation agent for epinephelus coioides sperm, which is an aqueous solution containing dimethyl sulfoxide and glucose, wherein the pH value is 7.5, the osmotic pressure is 328mOsmol/Kg, and each 1 liter of aqueous solution contains 100 milliliters of dimethyl sulfoxide and 60.1 grams of glucose. The invention also discloses an ultralow temperature cryopreservation method for the sperms of the Epinephelus coioides, which comprises the steps of mixing the semen with the cryoprotectant in equal volume, filling the mixture into a wheat pipe, placing the mixture in parallel at a position 3-5cm above the liquid nitrogen surface, staying for 8-10 minutes, and then directly putting the mixture into liquid nitrogen for preservation. The cryopreservation agent of the invention has simple preparation and low cost, and only has two non-reactive components, thereby reducing the adverse effect of the cryopreservation agent on sperms. The preservation method can make up the defect that no related preservation technology exists in the existing Epinephelus coioides.
