abstract | The invention discloses a preparation method of bazedoxifene, which comprises the following steps: a. adding a polar organic solvent, a non-proton Lewis acid organic solution and a hydrogen ion providing agent into 1- (4- (2- (azepine-1-yl) ethoxy) benzyl) -5- (benzyloxy) -2- (4- (benzyloxy) phenyl) -3-methyl-1H-indole, and stirring at 0-40 ℃ to obtain a reaction solution; b. and d, separating and purifying the reaction liquid obtained in the step a to obtain bazedoxifene. The method of the invention adopts non-proton Lewis acid such as boron trifluoride which is low in cost and easy to obtain as a catalyst for preparing bazedoxifene, has mild reaction conditions, convenient operation, good safety and low energy consumption, and the obtained bazedoxifene has high yield and high purity, reduces the preparation difficulty and the production cost of bazedoxifene, has very obvious positive effects, and is very suitable for being used in industry. |
