abstract | The invention discloses a method for visually marking a genomic locus. The invention creates a novel genome visual marker tool TTALE by fusing thioredoxin TRX at the C end of a traditional transcription activation-like effector element (TALE) protein. Experiments prove that: TTALE can be used to accurately label genomic repeats such as telomeres, centromere, and Ribosomal RNA coding sequences (Ribosomal DNA, rDNA), as well as coding loci (MUC4) in different types of human cells such as tumor cell lines, embryonic stem cells, adult stem cells, terminally differentiated cells, and the like. The TTALE technology can make up for the blank that the accuracy, simplicity and long-acting living cell genome visual marking technology is lacked in the current scientific research and clinical markets, thereby promoting the basic research of human aging and important diseases and the development of clinical diagnosis and treatment technology. |
