abstract The invention discloses a screening method for the unmarked deletion mutant of X. citri hfq gene, which comprises the following steps: (1) extracting the DNA of X. citri; (2) constructing a recombinant suicide plasmid vector with hfq gene deleted: A. ‑up‑F, Hfq‑up‑R, Hfq‑down‑F, Hfq‑down‑R respectively amplify the upper and lower flanking sequences of the hfq gene of X. citri; B, recover the PCR fragment; C, mix the recovered product with the carrier respectively pEASY‑T 1 vector ligation; D, the ligated products were transformed into Escherichia coli respectively; E, the positive cloned plasmid was extracted; F, the double enzyme digestion upstream/downstream T‑vector recombination plasmid, pK18mobSacB; DH5α Escherichia coli; (3) transforming the recombinant suicide plasmid vector into X. citri; (4) screening mutants by sucrose medium method.
