abstract | The present disclosure provides systems and methods for attaching nanopore-detectable tags to nucleotides. The present disclosure also provides methods of sequencing nucleic acids using the disclosed tagged nucleotides. Provided herein are tagged nucleotides and methods for attaching tags to nucleotides. The tags can be attached by chemical reactions such as "dick chemistry". In one aspect, the present disclosure provides tagged nucleotides comprising (a) a polyphosphate moiety with a terminal phosphate; and (b) via a triazole, 1,2-diazine, disulfide, secondary amine , hydrazone, thioacetamide, or maleimide-thio adduct covalently bound to the tag of the terminal phosphate of the nucleotide. |
