titleOfInvention | A method for detecting the influence on the secretory function of endothelial cells during the epithelial-mesenchymal transition of triple-negative breast cancer cells |
abstract | The invention relates to a method for detecting the influence on the secretion function of endothelial cells during the epithelial-mesenchymal transition of triple-negative breast cancer cells. The triple-negative breast cancer cell line MDA-MB-231 and human umbilical vein endothelial cells HUVEC are selected, and an experimental group is set up: MDA-MB-231 and HUVEC co-culture group, control group: MDA-MB-231 culture group alone; both experimental group and control group were stimulated with 10ng/ml TGF-β for 36 hours, and then quantitative proteome expression profile analysis by TMT The differential expression of secreted proteins in the cell supernatant between the two groups was compared, and the secretion results of SERPINE (PAI‑1) factor in the co-culture group and the single-culture group were compared. During the epithelial-mesenchymal transition of triple-negative breast cancer cells, triple-negative breast cancer cell lines can promote endothelial cells to secrete chemokine CCL5, thereby promoting the metastasis of triple-negative breast cancer cells. |
