abstract | The invention provides a biological preparation method of (1R,2S)-2-(3,4-difluorophenyl) cyclopropylamine D-mandelate (I). Specifically, the method of the present invention comprises the following steps: (a) in a liquid reaction system, using the compound of formula VI as a substrate, in the presence of a coenzyme, under the catalysis of carbonyl reductase, carry out an asymmetric reduction reaction, thereby forming the compound of formula V (b) the compound of formula V reacts with triethyl phosphonoacetate to obtain the compound of formula IV; (c) the compound of formula IV is subjected to aminolysis and Hoffmann degradation to form a salt with D-mandelic acid to obtain the compound of formula I. The present invention also provides a reaction system, comprising: (i) an aqueous solvent; (ii) a substrate, wherein the substrate is a compound of formula VI; (iii) a coenzyme; (iv) a carbonyl reductase; (v) a co-substrate and (vi) enzymes for coenzyme regeneration. |
