abstract | The invention provides a temperature-controllable electrochemical DNA biosensor and a preparation method thereof. The thiolated single-stranded DNA complementary to a target DNA sequence was immobilized on the surface of a gold wire thermode as a capture probe. After being complementary to the target DNA, a double-stranded structure with a 3' blunt end was formed to induce exonuclease III from the double-stranded structure. The 3' blunt end of the chain structure digests the capture probe from the 3' end to the 5' end, and the target DNA is released and undergoes a new hybridization Dicer cycle with other capture probes. Finally, the number of capture probes on the electrode surface is significantly reduced. The reduction of the electrochemical adsorption signal of the capture probe on the electrode surface before and after the enzyme digestion cycle to the detection solution of hexaammine ruthenium trichloride has a linear relationship with the logarithm of the DNA concentration, and the detection of the target DNA can be realized. The detection method provided by the invention has the characteristics of short detection time, high sensitivity, low detection line and good selectivity for the detection of target DNA. |
