The method for preparing the exopolysaccharide from the lactobacillus casei fermentation liquor comprises the steps of fermenting lactobacillus casei LC2W containing an NADH oxidase gene expression vector, and collecting the exopolysaccharide from the fermentation liquor, wherein the fermentation is aerobic fermentation. The method of the invention obviously improves the yield of extracellular polysaccharide, the activity of NADH oxidase and the yield of lactic acid in the fermentation process. The prepared extracellular polysaccharide has wide prospect in the application aspect of improving the immunity of medicines, health-care products or foods.
