abstract | The invention discloses an alkaline xanthine dehydrogenase and its application in a detection kit. The protein disclosed in the invention includes the large subunit of xanthine dehydrogenase and the small subunit of xanthine dehydrogenase. The alkaline xanthine dehydrogenase provided by the invention is the first reported xanthine dehydrogenase of Acinetobacter baumannii which has been confirmed by experiments. The alkaline xanthine dehydrogenase provided by the present invention can adapt to samples under alkaline determination conditions, has higher substrate affinity and the highest catalytic efficiency than the existing reports, and can degrade and detect hypoxanthine/xanthine more sensitively It can be combined with PNP enzyme and 5'-nucleotidase to detect inorganic phosphate and adenosine deaminase, which is beneficial to improve the catalytic efficiency of the enzyme, reduce the cost, and is more beneficial to the industrial application process. The present invention also provides methods of using xanthine dehydrogenase for the detection of xanthine/hypoxanthine content in samples including serum and urine. The present invention further provides a detection kit for detecting xanthine/hypoxanthine content in a sample. The detection method and detection kit provided by the invention are not affected by dissolved oxygen in the sample, have wider detection range and higher sensitivity than the prior art, and are convenient for popularization and application. |
