abstract |
The present application relates to isolated vertebrate cells and the like suitable for recombinant expression of a polypeptide of interest, wherein the vertebrate cell is altered to impair the function of an endogenous protease interstitial protease and wherein the cell comprises at least one heterologous protein encoding a polypeptide of interest Polynucleotides, wherein the polypeptide of interest is secreted by a cell. It was found that production of a recombinant polypeptide of interest using corresponding vertebrate cells significantly reduces cleavage of the polypeptide of interest secreted into the cell culture medium. Improved production and screening methods are also provided. |
