Methods and devices for long-read, label-free, optical nanopore long-chain molecular sequencing. In general, the present disclosure describes novel sequencing technologies based on the incorporation of nanochannels to deliver single long-chain molecules using wide-spaced (>wavelength), ~1-nm pore size "zigzag" nanopores that substantially slow down Translocations are used to provide massively parallel, single base resolution using optical techniques. A novel, directed self-assembled nanofabrication scheme using simple colloidal nanoparticles is used to form nanopore arrays on top of nanochannels, allowing long-chain molecules to unfold. At the surface of the nanoparticle array, strong localized electromagnetic fields in the engineered plasmonic/polarized phonon structure allow for single-base resolution using optical techniques. |
