The invention discloses a thioketal linking unit and its synthesis method and application; the structure of the thioketal linking unit is shown in formula (I): Wherein, R=-OH or -COOH; R 1 and R 2 meet one of the following conditions: (1) R 1 =R 2 =methyl; or, (2) R 1 =phenyl, p-methoxy phenyl, 2,4,6-trimethoxyphenyl, naphthyl or p-methoxynaphthyl, R 2 ═H; or, (3) R 1 and R 2 together form cyclohexyl or cyclopentyl. The fluorescein-labeled nucleotide obtained by linking the unit with nucleotide and fluorescein can be used for DNA sequencing. Compared with the prior art, when the reversible terminator of the linking unit of the present invention is used for DNA sequencing, its extension efficiency is close to 100%, and when the template is a plurality of consecutive identical bases, only one reversible terminator is extended at a time; its synthesis The required raw materials are simple and easy to obtain, and the synthesis process is a conventional chemical reaction, which can be used for large-scale popularization. |
