abstract | The invention discloses a new method for preparing acotiamide hydrochloride trihydrate (Acotiamide hydrochloride). The preparation method comprises the following steps: (1) taking 2-[N-(2,4,5-trimethoxybenzoyl) amino]-4-ethoxycarbonyl-1,3-thiazole (compound of formula I) , di-n-butylamine, N, N-dimethylformamide react to obtain 2-[N-(2-hydroxyl-4,5-dimethoxybenzoyl) amino]-4-(ethoxycarbonyl) ‑1,3‑thiazole (compound of formula II) crude product; (2) adding the crude product of compound of formula II obtained in step (1) into a mixed solvent of N,N-dimethylformamide and acetic acid to obtain compound of formula II (3) reacting the above-mentioned formula II compound with N,N-diisopropylethylenediamine to obtain a reddish-brown viscous liquid; (4) adding ethanol and concentrated hydrochloric acid, and reacting to obtain the crude product of acotiamide hydrochloride trihydrate (5) The crude product of acotiamide hydrochloride trihydrate was recrystallized to obtain refined acotiamide hydrochloride trihydrate.
