abstract | The invention discloses a construction method of a liver cancer cell strain capable of silencing FGFR4 gene expression and stabilizing liver cancer. The construction method comprises the following steps: firstly, aiming at the sequence of human FGFR4 gene, designing and synthesizing oligo target sequence of 3 pairs of optimal miRNA, respectively annealing the products and interfering vector pcDNA TM 6.2-GW/EmGFP is subjected to a connection reaction, escherichia coli is transformed, positive clones are selected, and sequencing verification is carried out. Secondly, the plasmid is transfected into FGFR4 high-expression cells, the expression conditions of various groups of FGFR4 in the mRNA and protein levels are detected through real-time fluorescent quantitative PCR and Western blot, and miRNA targets with the most obvious gene silencing effect on the liver cancer cell strain FGFR4 are screened out. After transfection, the gene can be used as a liver cancer cell line for stably silencing FGFR4 gene, and is used for further researching the gene function of FGFR4 in the occurrence and development of liver cancer. |
