abstract The invention discloses a method for determining content of effective substances in Yinmajiedu granules by an accelerated solvent extraction (ASE)-ultra performance liquid chromatography (UPLC), aiming at providing a method which is high in detection accuracy, time-saving and efficient, and can be used for determining the content of the effective substances in the Yinmajiedu granules by the ASE-UPLC. The method comprises the steps of (1) weighing 1g of Yinmajiedu granules, adding 10g of diatomaceous earth into the weighed Yinmajiedu granules, and evenly mixing for later use; (2) adding the mixture obtained in the step (1) into an extraction pool, wherein a filter membrane is put into the extraction pool in advance, adding the diatomaceous earth onto the mixture until reaching a mouth of the extraction pool, covering the extraction pool with a cover, then carrying out static extraction, and obtaining an extracting solution after extraction; (3) transferring the extracting solution into an evaporating dish, evaporating the extracting solution to dryness and then dissolving by using methyl alcohol, determining the volume of the product in a 10mL of volumetric flask, absorbing 1ml of sample liquid, adding the absorbed sample liquid into a 2ml of centrifuge tube filled with a graphite carbon solid phase extraction (SPE) column for purifying, filtering the solution by a 0.22-micron filter membrane, and then injecting filtrate into a UPLC for determining. The method for determining the content of the effective substances in the Yinmajiedu granules by the ASE-UPLC belongs to the technical field of chemical detection.
